Oxford nanopore sequencing

Oxford Nanopore Technologies (ONT) is a provider of long-read sequencing technology that offers real-time analysis of DNA and RNA molecules. The ONT platform utilizes nanopore-based technology that allows for direct sequencing of single DNA or RNA molecules, resulting in long reads that can span entire genomic regions, including complex and repetitive regions that are difficult to sequence with other technologies. The long read lengths and real-time data analysis capabilities of ONT sequencing make it a powerful tool for a wide range of applications, from de novo genome assembly and structural variant detection to transcriptomics and epigenetics research.

In CFG, we offer full sequencing service on the P2 Solo platform, that uses Promethion flowcell and enables running up to two flowcells in parallel. The kits used in library preparation can be discussed with the client. We currently provide service with Kiit14 chemistry – Ligation kit SQK-LSK-114,  rapid barcoding SQK-RBK-114, Rapid seequencing RAD114, native barcoding NBD114 and 16S sequencing. Other kits are not routinely used, but their application can be arranged after discussion with the client

Oxford Nanopore platforms offer Adaptive Sampling, a unique way of target enrichment that uses real-time basecalling, alignment and pore control to selectively enrich or deplete sequences. Average enrichment depends on a proportion of th target/offtarget sequence in the sample, but can generally reach 5X to 50X enrichment. If the customer requires to employ Adaptive sampling, a .fasta file (and optionally a .bed file) needs to be provided.
